Born in Douglas, County Cork, Murphy initially hoped to work as a musician, writing songs and playing guitar with various bands, before joining Corcadorca Theatre Company and appearing in Disco Pigs. Murphy appeared in a number of independent films and plays including The Seagull and Much Ado About Nothing before his first mainstream success in 28 Days Later.

Murphy played a wide of roles in his career, from those in his native Ireland such as Intermission and The Wind That Shakes the Barley (both of which became the highest grossing Irish films to date) to more blockbuster fares like his role as Dr. Jonathan Crane in The Dark Knight Trilogy. He also appeared in the popular drama series Peaky Blinders.

Murphy twice sang onscreen, firstly in Disco Pigs in which he also contributed the song "So New" to the soundtrack, and secondly in Breakfast on Pluto, in which he sang "Sand."
